Development

CA vector containing the human IL-6 gene downstream of the CMV promoter was microinjected into (NOD♀ x NOG♂) F1 pronuclear stage embryos, and crossed with NOG mouse to establish them.

Product features of Humanized NOG-IL6 mouse
  • HSC engrafted NOG-IL6 (hereafter: huNOG-IL6) mouse exhibited enhanced human monocyte/macrophage differentiation.
  • Tumor-infiltrating macrophages of huNOG-IL6 mouse expressed high levels of CD163, a marker of immunoregulatory myeloid cells, and produced immunosuppressive molecules such as arginase-1 (Arg-1), IL-10, and VEGF.
  • Such cells from huNOG-IL6 mouse, but not those from conventional huNOG mouse, suppressed the proliferation of human T cells in response to antigenic stimulation in in vitro culture.
  • These results suggest that functional human TAMs can be developed in NOG-IL6 mouse and are expected to contribute to the development of new cancer immunotherapies targeting immunomodulatory/immunosuppressive myeloid cells.
